Granulation Endpoint Detection Case Study

Wet granulation is still an important process for tablet manufacturing and we are always looking to add new predictive methods to sit alongside compaction simulation.  

We are thrilled to announce a collaboration with Caleva to offer predictive tests at Merlin to support wet granulation. We can help small scale lab granulation through the use of the Caleva MTR.

The Mixer Torque Rheometer (MTR) was used to investigate the quantity of water required to granulate using the Binder addition function. Then the effect of mixing time was investigated to assess the robustness of the formulation.

Set Up

A 100g batch was used for the investigation. The blend was weighed into an amber glass jar and blended for 10 minutes using a Turbula T2 blender at 33rpm. The MTR chamber holds around 50 ml volume of powder. The formulation was transferred to a measuring cylinder and 50 ml (~20g mass). 

The first test assessed the change in the formulation behaviour with increasing amounts of granulating fluid. The binder was present in dry form in the blend and water was used as the granulating fluid. 

The next step of the investigation looked at sensitivity to mixing time. Different water addition volumes were used.

Conclusions

Using a small batch size, the investigations have been able to successfully assess the maximum granulation fluid proportions for the formulation.

The mix time experiment shows that higher liquid to solid radios, produce a granule with good levels of torque which is also maintained during extra mixing. 

With lower liquid to solid ratio the results showed under granulation. The low torque showed the formulation will not produce granules with sufficient strength. For this formulation the level of granulating fluid is more important than the mix time.
